Vacuolar transport of glucosinolates was performed with vacuoles isolated from Brassica leaves, barley and Arabidopsis cell cultures. Isolation and purification of vacuoles from Brassica is extremely difficult. The two experiments, which could be performed with Brassica vacuoles, indicate that in Brassica glucosinolates are taken up by a proton/glucosinolate antiport system. In contrast, barley vacuoles use a direct energized transport system to take up glucosinolates. In order to have a system where vacuoles can be isolated more efficiently from a plant producing glucosinolates we developed an isolation procedure starting from Arabidopsis cell cultures. Surprisingly glucotropaeolin, our radiolabeled glucosinolate, uptake in vacuoles isolated from cell cultures was directly energized as observed for barley. This may be due to the fact, that the Arabidopsis cell culture produce no or only very low amounts of glucosinolates. In order to verify whether glucosinolates are transported by a proton antiport or direct ATP dependent manner, in glucosinolate producing plants, we have now established a vacuole isolation procedure for Arabidopsis leaves and in the near future we will investigate glucosinolate transport using these vacuoles.
