Abstract

I have carried out the following studies, which in aggregate address all subtasks and deliverables of WP5:

1. Established a fully annotated list of ~1000 genes involved in metabolic, signalling and differentiation pathways controlling glucose homeostasis.
2. Tested oligo (50-mers) microarrays and different labelling techniques for cRNA preparations and hybridization. Chose not to further use long oligo arrays.
3. Generated ~630 cDNAs from the gene list mentioned in 1. by RT-PCR, subcloning and sequence verification. These cDNAs have been amplified and have been printed as microarrays on glass slides.
4. Generated a cDNA library from fat obtained from control, obese and diabetic mice. Sequenced ~1000 individual clones representing 550 different genes, of which 77 are not represented in publicly available EST data bases. These cDNA will complete the list of genes to be printed on microarrays.
5. Generated a cDNA library from subcutaneous and visceral fat obtained from control, obese and diabetic human patients.
6. Completed the metabolic characterization of genetically identical mice under high fat diet and which develop different phenotypes: obesity with diabetes, no obesity but diabetes and no obesity nor diabetes. Stored liver, muscle, adipose tissue, hypothalamus from large number of these mice for microarray analysis.
