In depth-knowledge of the three different dPCR systems including the software will be built. The comparability of the three systems will be investigated using GMO reference materials that are available in the NRL and are routinely used. As an alternative or in addition to the GMO reference materials, the Vibrio-Sequin DNA standards (Flütsch et al., 2023) with the corresponding assays can also be measured on the three devices in order to obtain a comparison. Suitable protocols for the three systems will be developed, taking into account the technical specifications of the devices (permissible master mixes and probes) and, as far as possible, the existing standard protocols. Finally, the measurements obtained will be statistically evaluated, including measurement uncertainty.

Projektziele

The aim of this project is to gain in-depth knowledge of three existing digital PCR (dPCR) systems (Stilla Naica, Qiagen QIAcuity, Bio-Rad CX200) and to investigate their comparability and the reproducibility of measurements. The suspicion, based on previous practical experience, is that these systems deliver different results although they should measure absolute quantities.
