COST-Action BM1307 - European network to integrate research on intracellular proteolysis pathways in health and disease

Proteolytic events regulate a myriad of cellular responses, ranging from cell signaling to protein recycling. Our laboratory has shown that limited proteolysis of the p120RasGAP protein by proteases of the caspase family plays an important homeostatic function in stressed cells. While this work showed that only a subset of caspase substrates are cleaved in stressed cells, the overall landscape of proteolysis in cells subjected to non-lethal forms of stresses is not known. The goal of this project is therefore to generate, using mass spectrometry state-of-the-art technologies, a global map of the proteins that are cleaved in response to various forms of non-cell death-inducing cellular stress and to assess the contribution of specific proteases in these proteolytic events. Such global proteolytic profiling associated with stress is in line with the objective of the PROTEOSTASIS cost action and the outcome of these analysis will be shared with our PROTEOSTASIS partners. The data generated by this project are expected to contribute significantly in the characterization of signaling pathways regulated by stress in cells and consequently in the required information to establish strategies to modulate these pathways in order to dampen the negative consequences associated with cellular stress.
