Nagetiere werden am Ende von Tierversuchen oder als Futtertiere für Zoos euthanasiert. Es gibt verschiedene Methoden dies zu tun und die Bedenken betreffend Tierschutz und dem Stress, der die Euthanasie für die betroffenen Tiere bedeutet ist dabei unterschiedlich. Der Gebrauch von CO
2
führt zu grossem Stress und Fluchtverhalten. Inerte sowie Edelgase sind mögliche Alternativen, da sie keinen stechenden Geruch aufweisen und bei subanästhetischen Dosierungen kein Fluchtverhalten auslösen. Wir haben vor kurzem bei Mäusen gezeigt, dass Euthanasie ausgelösst durch ausschliessliche Hypoxie mit Stickstoff weniger Stress bedeutet als wenn CO
2
eingesetzt wird. Das vorliegende Gesuch will dies weiter untersuchen. Es sollen verschiedene Mausstämme im eigenen Käfig oder eigens dafür gebauten Gefässen mit Stickstoff euthanasiert werden. Im ersten Teil werden EEG, Bewegung, Fluchverhalten, „Grimace scale“, und Korticosteron spiegel gemessen bei Individuen gemessen wie in unseren bisherigen Experimenten. Im 2. Teil werden grössere Gruppen (20-30 Tiere) in Zoos evaluiert und auf Video aufgenommen. Bewertet werden die Bewegungsaktivität und Lautäusserungen. Ziel dieser Untersuchung ist es eine Euthanasiemethode zu finden die sicher ist für involviertes Personal, erschwinglich und mit minimalem Stress für die betroffenen Tiere. Es wird erwartet, dass die Ergebnisse der Studie zu einem Umdenken bezüglich CO
2
Euthanasie führen und das Wohlerhgehen der Tiere in Zoos und in Versuchseinheiten verbessert werden kann.

Rodents are commonly euthanased following the end of experimental procedures and for use as food animals in zoos. There are various methods employed for euthanasia, all of which have welfare concerns regarding the level of stress and pain/nociception caused to the animals. Carbon dioxide euthanasia is associated with a large degree of stress and fleeing behaviour. The Nobel and other inert gases offer an attractive alternative since they are non-pungent and do not elicit a fear response at sub-hypnotic levels. We recently demonstrated that euthanasia by hypoxia using nitrogen gas alone is less stressful for mice compared to carbon dioxide. This project aims to evaluate further the use of nitrogen gas for euthanasia in differing mouse strains in both purpose built gas-tight chambers and also the home cage. Phase I of the project will measure EEG, locomotion activity, behavioural fleeing responses, grimace scale and plasma corticosterone of individual animals as in our previous experiments. Phase II will evaluate gas euthanasia of cohorts of mice in zoos using state of the art video tracking for locomotion activity and ultrasonic vocalisation. These mice will be euthanased in the same cohorts as the home cage and recordings will be made in either the home cage or purpose built gas chamber. The ultimate aim is to find a euthanasia technique, which is affordable, safe for the technicians involved and offers the minimal amount of stress to the animals used. It is hoped that the findings will refine euthanasia techniques in research facilities and zoos to improve animal welfare.

The principle aims of this project are to comparatively quantify stress in rodents using a number of modalities; namely EEG, locomotion, grimace scoring, vocalisation and hormonal assays during gas euthanasia. The ultimate aim is to determine a methodology by which mice may be euthanased with the minimum amount of stress while maintaining personnel safety. The data is to be disseminated in international journals and conferences to encourage the refinement of stress free euthanasia of rodents in both research facilities and zoos.
