Targeting protein-protein interactions with macrocycles is gaining ground in drug discovery as novel therapeutic options. Stapled peptides have emerged as powerful tools to target these interactions. However, the development of stapled peptides relies on cumbersome screening. Here, we wish to use synthetic biology to develop a novel technology to generate billions of stapled peptides in a test tube, and screen them against any target with no need of structural information.
