Development of a predictive model using porcine intestinal mucosa to evaluate drug absorption in the gastrointestinal tract and so determine the effects of active transporters, metabolism and formulation excipients. The aim is to enable early identification of lead candidates and elimination of poor candidates to optimise financial and human resources. Existing models include artificial membranes, cell cultures (e.g., Caco-2) or rat intestine; these are inadequate predictors of uptake in man, not physiologically realistic and ethically questionable.
