For the regeneration of soft tissues, poly(4-hydroxybutyrate) (P4HB) produced by microorganisms is one of the most promising scaffold materials. Furthermore, P4HB-based sutures have been approved by the FDA for clinical application. However, a systematic study on the production process, downstream process (DSP) and fabrication of P4HB for tissue engineering has not yet been reported, although such processes are imperative for the application and cost management of a material in medical fields. Especially, gaining access to medical-grade P4HB is highly important for tissue engineering of heart valves in Switzerland, and plays a critical role for the success of the startup company Xeltis. Within this project we develop sustainable and reproducible processes for production and purification of P4HB and will apply the material in the fabrication of scaffolds for tissue engineering. We expect to significantly reduce P4HB production costs and accelerate the way to GMP production of P4HB for clinical studies and, most importantly, to enhance the competitive advantages of Xeltis AG.
