Abstract

We have recently discovered new peptidomimetic antibiotics targeting a protein (LptD) essential for outer membrane biogenesis in Pseudomonas spp. The peptidomimetics protect against P. aeruginosa in a mouse infection model, and a clinical lead has entered a phase I clinical trial. The goals of this project are to extend the spectrum of activity of these compounds, and to develop new antibiotics targeting other Gram-negative pathogens. The work should lead to an important new class of antibiotics to combat drug-resistant Gram-negative 'super-bugs'.
