Laccase-mediator systems (LMS) are increasingly recognized as valuable tools for mild oxidations in organic synthesis. Numerous fragrance compounds are alcohols, aldehydes or ketones derived from unsaturated hydrocarbons (terpenes). In this project improved LMS suitable for mild oxidations of terpenes and other fragrance precursors will be developed. Novel laccases of bacterial and/or archaeal origin will be expressed in Escherichia coli and improved by directed evolution using novel colorimetric screening tools. The evolved LMS will be used for fragrance synthesis from pure compounds and complex substrate mixtures in the mL to L scale in order to find novel fragrance notes (new chemical entities) and innovative and sustainable production methods for fragrances.
