Abstract (Englisch)	The discovery of RNA silencing has opened up a completely new avenue for therapy. The main goal of this project is to determine the structure of the dsRBDs of three proteins which are part of RISC (RNA Induced Silencing Complex), namely Dicer, TRBP (TAR RNA Binding protein) and PACT (Protein Activator of PKR), in their free state and in complex with high affinity siRNA and pre-miRNA. The structure of these dsRBD-RNA complexes will be used to design new siRNA- and miRNA-based therapeutics, which will eventually be tested in in vivo mouse models
