Development and validation of highly predictive anticancer compound screening (HIPACS) systems: impact of tri-dimensional structure and stromal cells on sensitivity of human tumors to drug treatment.
Résumé des résultats (Abstract)
(Anglais)
Screening of anticancer drugs is currently performed on established human tumor cell lines cultured in monolayers. Predictive value is modest as shown by low success rate of compounds entering clinical studies possibly due to cell selection within permanent cell lines and to absence of tridimensional (3D) structure or tumor-stroma interaction. In this project advances in the 3D culture of primary cancer cells and mesenchymal stromal cells (MSC), and progress in cell imaging technology will be used to establish novel platforms for anticancer drug evaluation.
