Expression and characterisation of recombinant human collagen (COMA)
Abstract
(English)
In ubiquitous use, collagens are extracted and purified from natural sources. The non-uniformity of such raw materials, being composed of several different collagen types, and safety concerns hamper their use for biomaterials or pharmaceuticals. To overcome these obstacles, recombinant technology will be trialled. Samples of rh-procollagen (with intact triple helix structure) to be used for prototyping new medical materials (e.g. synthetic tissues) will be gained from high-cell-density processes with a yeast expression system. Moreover, the experimental data collected will provide first estimates of manufacturing costs in terms of (i) raw materials and (ii) capability figures for the cultivation, downstream and further biosynthetic processing needed to achieve the mature functional form required.
