The project aims to construct recombinant bacterial vaccine strains expressing the epidemiologically most relevant colonization factors of enterotoxigenic Escherichia coli (ETEC) in view of producing a vaccine strain against enterotoxigenic E. coli infections. The primary purpose of the project is to construct strains expressing the desired antigens in a correct conformation in order to ensure best possible immunization by the vaccines. Besides genomics and proteomics, particular importance will be attached to the structural aspects by implementing fluorescence- and electron-microscopy in combination with optimal gene expression analyses. For assessment of the stability of the recombinant vaccine strains, specifically designed gene micro-arrays will be developed. The recombinant strains will be assessed for medical efficacy and biological, medical and environmental safety.
