COST-Action D28 - Natural Products as a Source for Discovery, Synthesis and Application of Pharmaceuticals

Within a network dedicated to the synthesis of biologically active alkaloids, this project aims at the synthesis of (+)-Vertine. This natural product of the Lythracea alkaloid family plays a role in glucose level regulation in blood and lowers blood pressure; it is a sedative and has antiinflammatory properties. Vertine has not been synthesized previously. Its interesting pharmacological properties apart, the synthesis of the 13-membered lactone incorporating a Z alkene, a diaryl unit and a hydrophyridine is synthetically challenging and will present opportunities to develop and test new synthetic methods. Testing the biological activity of the target molecule and of some analogues will be possible within the network as well as via individual industrial collaborations.

(+)-Vertine is a member of the Lythracea alkaloid family with a number of interesting biological activities, notably glucose level regulation in blood and blood pressure reduction. Vertine has not been synthesized previously. Its interesting pharmacological properties apart, the synthesis of the 12-membered lactone incorporating a Z alkene, a diaryl unit and a hydropyridine is synthetically challenging. The question of an induced chiral axis of the biaryl unit by the other stereogenic elements in the constrained macrolactone molecule is intriguing.
