Non-conform DNA content as a marker of somaclonal variation in potato The cytometric investigation of plantlets regenerated from potato internodes revealed that all the phenotypically non-conform plants had in their cells nuclei an abnormal content of DNA. But also a few apparently normal looking regenerants presented such nuclei with aberrant DNA content. None of the regenerants that had normal tetraploid nuclei exhibited phenotypic differences compared to micropropagated, non-regenerated plants. No relation could be established between the type of cytometric profile and the type of the observed phenotypic abnormalities. The conclusion that could be drawn from these observations is that the regenerants having nuclei with abnormal DNA contents arose from cells which underwent erroneous mitosis. During this event, the duplicated chromosomes were unevenly distributed in each of the daughter cells. This phenomenon of uneven mitosis affects for each regenerant different chromosomes, hence comes the large diversity of phenotypes.
