The Ubiquitin-proteasome pathway is a very attractive target for pharmacological intervention because it is necessary for the regulated and selective degradation of intracellular proteins involved in the control of most biological processes, including proliferation, and alterations of its activity or selectivity often contribute to human pathologies, especially cancer, inflammatory and degenerative diseases.

However, due to the complexity of this pathway and to the variety of its substrates, major advances towards the design of drugs capable to alter the stability and thus the level of one or a few pertinent protein(s) require a deeper understanding of the fine tuning of its activity and selectivity. This project proposes an integrative analysis of the multiple biochemical, structural, spatial and temporal parameters that determine the degradation of ubiquitous regulators of cell proliferation, and to design novel anti-cancer strategies and molecules.
